Pagoclone is an anxiolytic drug belonging to the class of non-benzodiazepines of the cyclopyrrolone family, an analogue of the sleeping drug zopiclone, developed by the French team Rhone-Poulenc & Rorer S.A..
Studies in rats have shown that pagoclone acts on GABA receptors (0.7-9.1 nM), and is a partial agonist of α1-, α2- and α5- GABA receptors, as well as a complete agonist of α3-receptors.
This drug is not a benzodiazepine, as it has a completely different chemical structure, but it can cause a similar sedative and amnesic effect.
It does not cause side effects such as aggression, amnesia, loss of coordination, nausea, less damage to the liver. Its effect can be quickly reversed with the drug flumazenil (antidote).
Pagoclone never went on sale, due to fears of abuse, and as a result, research on its use was discontinued.
