Description

2-Fluorodeschloroketamine(2-FDCK), 2′-Fl-2-Oxo-PCM) is a dissociative drug, an anesthetic that has a similar structure to ketamine and is its analog with the difference that the chlorine group is replaced by fluorine, known as a designer drug.

History

The synthesis of 2-FDCK was first described in 2013 as part of the study and synthesis of new anesthetic drugs and ketamine analogues. In the designer drug market, 2-FDCK was mentioned by EMCDDA in 2016

Structure

2-FDCK or 2-(2-fluorophenyl)-2-(methylamino)cyclohexan-1-one.
Preparation of 2-FDCK from fluorobenzonitrile belongs to the class of arylcyclohexylamines, which includes other well-known drugs such as ketamine, PCP and other derivatives. Structurally, 2-FDCK consists of a cyclohexylamine unit with an aryl group attached to the same carbon as the amine. The o-fluorophenyl group is used as the aryl substituent and the amine group is methylated, the cyclohexyl ring has a ketone group next to the amine.
2-FDCK differs from ketamine in that the phenyl group has a fluorine atom attached to it, whereas ketamine has a chlorine atom.

Synthesis

The order of synthesis of 2-FDCK can be divided into a five-step reaction. 2-Fluorobenzonitrile enters into a grignard reaction with cyclopentylmagnesium bromide, after a bromination reaction, the reaction of the resulting ketone with methylamine, producing a water-soluble HCl salt.

Pharmacodynamics, dosage

2-FDCK has a structure similar to ketamine, but the effects of the drugs are not much different because the halogen in position 2 is not chlorine but fluorine, and the molecule is less polar. 2-FDCK is still poorly understood and little is known about its pharmacological and toxicological properties.

Legal status.

2-FDCK positions itself as a designer drug, but it is already banned in Italy, Japan, Latvia, Singapore, Sweden, Switzerland, Canada, Belgium, and the United Kingdom.
